CD37 and CD44 evaluation by flow cytometry: can these markers improve B cell lymphoma characterization?

چکیده
Diffuse large B cell lymphoma (DLBCL) is a heterogenous entity with many prognostic ancillary tests that are based on immunohistochemistry. These include cell-of-origin determination by the Hans criteria and “double expresser” characterization. Additionally, MYC fluorescence in situ hybridization required to distinguish DLBCL from its morphologically similar counterpart, hit lymphoma” or high-grade (HGBCL), “Burkitt-like” worse prognosis than DLBCL. All such require judicious triage of oftentimes limited specimens. Loss expression CD37 has been associated outcomes Absence CD44 more commonly seen Burkitt Very few studies have evaluated flow cytometric analysis, which permits quantitative assessment. To evaluate utility these two markers, we retrospectively reviewed seventy-three non-Hodgkin specimens was cytometry. Median intensity ratio (MFIR) calculated dividing median population background non-B mononuclear cells. In lymphomas DLBCL-like morphology, loss rearrangement (MFIR rearranged 0.3082 vs intact 1.09, p=0.003). also germinal center cell-like (GCB) GCB 0.4139 non-GCB 1.499, p=0.036) as other studies. DLBCL, not cell-of-origin, double expresser status, rearrangement. While our findings confirmation larger case series, decreased cytometry an otherwise biopsy features may provide impetus for FISH testing ultimate classification.

Tetraspanin CD37 protects against the development of B cell lymphoma.
Worldwide, B cell non-Hodgkin lymphoma is the most common hematological malignancy and represents a substantial clinical problem. The molecular events that lead to B cell lymphoma are only partially defined. Here, we have provided evidence that deficiency of tetraspanin superfamily member CD37, which is important for B cell function, induces the development of B cell lymphoma. Mice lacking CD37...

We studied 48 non-Hodgkin lymphoma (NHL) fine-needle aspiration (FNA) specimens with initial cytomorphology (CM) and flow cytometry (FC) and subsequent surgical biopsy of the same lesion to determine whether a reliable diagnosis of large cell lymphoma or large cell transformation could be made. CM was evaluated by examining 200 lymphocytes in each specimen. FC was performed by analyzing monoclo...

Background Cancer stem cells play crucial roles in resistance to therapeutic schemes and relapse of disease, so it is important to find targeted therapies that kill them selectively. Breast cancer is the most common cancer in females living in all part of the world including Iran and it has an important burden in public health with direct impact on patients’ families. Breast cancer in young ad...
متن کاملAssessment of CD37 B-cell antigen and cell of origin significantly improves risk prediction in diffuse large B-cell lymphoma.
CD37 (tetraspanin TSPAN26) is a B-cell surface antigen widely expressed on mature B cells. CD37 is involved in immune regulation and tumor suppression but its function has not been fully elucidated. We assessed CD37 expression in de novo diffuse large B-cell lymphoma (DLBCL), and investigated its clinical and biologic significance in 773 patients treated with rituximab plus cyclophosphamide, do...
متن کاملB cells with high side scatter parameter by flow cytometry correlate with inferior survival in diffuse large B-cell lymphoma.
Despite advances in the understanding of diffuse large B-cell lymphoma (DLBCL) biology, only the clinically based International Prognostic Index (IPI) is used routinely for risk stratification at diagnosis. To find novel prognostic markers, we analyzed flow cytometric data from 229 diagnostic DLBCL samples using an automated multiparameter data analysis approach developed in our laboratory. By ...
